Analysis Of Andrographolide Compound From Sambiloto Plant (Andrographis Paniculata Nees) On Alpha Glucosidase As Type II Antidiabetes

Abstract

The bitter plant contains the Andrographolide compound which can bind to the Alpha Glucosidase protein which can be analyzed by molecular docking using various websites and applications including pubchem, pass online, pharmMapper, swiss targetprediction, string db, swiss-model, fasta, Saveswebserve, pyrix PyMOL, protein plus, swissadme and protox II. The purpose of this analysis is to determine whether the Andrographolide compound is present with the target protein, namely alpha glucosidase, as a type II antidiabetic. Based on the results of this analysis, it was found that there was a hydrophobic bond and there was Met122A which indicated the target protein. This interaction indicates that andrographolide can inhibit alpha glucosidase so that not all oligosaccharides or polysaccharides can be broken down into monosaccharides, so it can prevent and stop the development of type II diabetes with a binding affinity result of -7.6 kcal/mol, this indicates that the more negative the binding affinity results, the natural compound. is the best compound. However, the toxicity level obtained was LD50: 1890mg/kg body weight. Based on the analysis results, it was found that the Andrographolide compound was included in class 4 with LD50: 1890mg/kg, meaning it was dangerous if swallowed because it was hydrophobic and difficult to dissolve in water.
