Abstract
This research aims to know the morphometric and genetic characters of fruit fly Bactrocera dorsalis complex that originated from the citrus plantations that be found in Simalungun district, North Sumatera province. This investigation goals also in understanding the relationship among body length of fruit flies and its other morphological characters. The sampling of fruit flies has been done on the citrus crops that located in Baringin Raya village. The fruit flies were collected by using steiner traps that be given with methyl eugenol lure on cotton. The fruit fly identification was carried out by morphology and DNA barcoding approaches. The morphological characters that be observed under Stereo Zeiss-Stemi 2000-C microscope and measured were body length (Y), wing length (X1), wing width (X2) and tibia hind leg length (X3). DNA barcoding working was done by isolation, amplification, sequencing steps and blasting by using MEGA 11 software. Morphometric traits and its relationship were analyzed by helping of software IBM SPSS Statistics 23. The result of investigation showed that the fruit flies that be collected from Simalungun distric citrus crops based on morphology and be confirmed by DNA Barcoding (mt COI) approaches belong to Bactrocera dorsalis complex. The mean of the body length, wing length, wing width, and tibia length of obtained fruit flies Bactrocera dorsalis complex (n=30) were 6.91, 6.36, 2.43 and 1.98 mm, respectively. The multiple linier regression equation among body length of fruit fly (Y) (n=30) toward its other morphological traits (X1-X3) was displayed by equation Y = 0.24 X1+ 0.58 X2 + 3.91 (R=0.85). The length of mtCOI DNA sequence of obtained fruit fly was 690 bp and dominated by Pyrimidine concentration 50.5%. The concentration of T(U), C, A and G nucleotides in the mtCOI sequence were 32.50%, 17.90%, 31.50%, and 18.0%, respectively.
